I’m newly practicing tissue culture 2 months ago. And I’m wondering if the initiation stage of banana tissue culture needs really hormones. For the initiation explant should first start the vegetative growth and I guess that within the second step of shooting hormones are necessary.
Some one can correct me.

Some put excised shoots directly on multiplication medium, so initiation stage is not separately.
If you want it to be separately, you may do not need growth regulators during initiation, but you should add antioxidants like ascorbic acid / cysteine to reduce blackening of tissue.
